Offering PacBio services on the newest PacBio Sequel II as of June 2019, all Sequel I systems have been retired. The Sequel II system produces 8-10x the data amount for ~2X the cost of the Sequel I with the same quality profile.
We can take samples/libraries from any stage and carry them through to sequencing. We offer High Molecular Weight Extractions, PCI cleans for user provided gDNA, shearing, BluePippin and ELF size selection, low input library preps, HiFi (12kb ccs) Library preps, Long Insert library preps up to 100kb, and 8M SMRTcell processing. We can demultiplex and generate ccs reads as well but further analyses cannot be accommodated in house and will be outsourced to UC Davis.
The Sequel II system generates about 6M reads worth of data per SMRTcell, ~80-100Gb of long insert (CLR) data or ~300Gb of HiFi rolling circle data (15-20Gb Unique Molecular content after ccs and filtering for 99% accuracy).
CLR genomes generally require ~65X coverage and HiFi assembles require ~30x.
Full length Transciptome sequencing or IsoSeq can also be performed for the same cost as the normal SMRTbell Library prep. Multiplexing can be accommodated. A human sized transcriptome would require 1 8M SMRTcell unless very rare transcripts are required and then 2 SMRTcells are recommended.
